Yes and no. Although the film is about Susanna Kaysen and her stay at a mental hospital, there are many parts of the film that do not hold true to the non-fiction work of Kaysen. One of the more notible inaccuracies in the movie was when Susanna and Lisa escaped from the hospital and went to Daisy's house on their way to Disneyworld. This escape never actually happened and Daisy's suicide was done at her new apartment but Susanna and Lisa were not there, they were in the hospital, however, how she did it remained unknown to the patients. Also, Susanna's stay at Mclean (the real name of the mental hospital) was a bit longer than what was shown in the film.
Again, yes and no. Angelina Jolie has been noted as saying that it was hard for her to become close to Winona Ryder because if she had seen her in pain in the mornning say with a headache, then screaming at her in a scene later that day would be very difficult. For the role and her character's sake, she could not look at the other actresses as whom they actually are. However, when the film was over, all of the girls had a special bond. Some noted friendships are that of Angelina Jolie and Jillian Armenante as well as Winona Ryder and Brittany Murphy who went on to do a one night play together on Broadway.
